The use of the poly(styrene-co-maleic acid-co-X%(N-benzyl)maleimide) copolymers enables 100% detergent-free stabilization of membrane proteins. These polymers act like molecular "cookie cutters," facilitating direct membrane solubilization and stabilization in a single step directly from the cell membrane, while preserving proteins in a near-native lipid environment.
Nanosene’s BzAM, produced through advanced polymerization techniques, is a well-defined polymer with a molecular weight (Mw) of ~5,500 g/mol and a low dispersity (narrow chain length distribution), a significant improvement over previous SMALPs. The BzAM series (BzAM25/BzAM30/BzAM35/BzAM40) is a highly tunable series of polymers designed for systematic and methodical research. This series enables researchers to screen for their target protein before scaling up their assay and to investigate structure-activity relationships across various protein types and membrane systems.
One of the most exciting features of the new BzAM polymers is the unique tunable platform they provide for investigating polymer amphiphilicity in target protein extraction. Research has shown that not all polymers are effective for all protein types, and this series enables the evaluation of the amphiphilic parameters required for optimal protein solubilisation from different membrane environments.
This product is compatible with various phospholipid types, including saturated lipids, e.g., 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC), and unsaturated lipids, e.g., 1-palmitoyl-2-oleoyl-glycero-3-phosphocholine (POPC).
The BzAM-membrane protein complex is suitable for a wide range of biophysical assays, including SDS-PAGE, SEC, Western blot, UV/Vis spectroscopy, and various chromatographic techniques, while compatible with affinity purification strategies.
